Thanks for all the responses! No, I wasn't aware of the official Toilet Map, so thanks to everyone who linked it! And cheers for the food ones, too; I didn't realise a few of the stations had cafés, bakeries, etc.!

If you are going between Kenton and Northwick Park on foot, there is a small tuck shop-type stall on the northside doorway at Northwick Park that is conveniently sited for quick use and the right direction to/from Kenton.

There are toilets on the platform at Preston Road, which (if your route is planned in such a way) is a good point where you might double-back while clearing the north-east chunk of Metropolitan Line. Usefully, both eastbound and westbound platforms are on the same platform island, with the toilets housed in the middle.

Moor Park has toilets on the platform similar to Preston Road

On one of my recce runs, I missed a very tight connection at Preston Road because the toilet block was in the way!

Nothing wrong with that as a first attempt - I think one of Andi's early world records was 17:56. You'll learn lots from doing it.

Toilets on the Underground are universally disgusting, but I seem to remember W Ken and Rayners Lane being especially foul. Signage is often rather discreet. Be aware that LU locks its loos at 9pm! The toilets at Wimbledon are NR and therefore well kept but cost to get in!

As regards eating and drinking, I think the key thing is giving yourself permission to stop.The first time I did the challenge, I was always to anxious about making the next train and therefore didn't eat or drink enough. I suffered for it the next day. Being a country bumpkin, I was surprised that London shops don't seem to open especially early. Besides the ones others have mentioned, I can think of:
newsagents type Kiosk on Morden station
Newsagents just to R of Canons Park
Gregs to R of Edgware stn
Open all hours type just outside the barriers on the shuttle side of Finchley C (if you've a wait for a shuttle)
Open all hours on road from Kenton towards Northwick Park
And seemingly NO late night takeaways in Amersham! Boo!

waiting for the 20 min hainault loop Leyton has an toilet on platform

Woodford westbound platform has toilets and snack hut. Both very useful.
